ABSTRACT The elephant grass (Cenchrus purpureus (Schumach.) Morrone) is one of the species with great prominence for energy production because of its favorable traits, such as high yield and short cycle, combined with several biomass quality characteristics. Therefore, this study intended to evaluate the morpho-agronomic traits of eight elephant grass genotypes under different cutting ages. A randomized block design with three replicates was applied to the experiment, and plots were composed of a 12 m long line divided into six subplots for each of the bimonthly cuts. Eight genotypes were evaluated in the plots, and the cutting ages (2, 4, 6, 8, 10, and 12-mo) were analyzed in the subplots. The following traits were assessed: DM yield, (DMY) percentage of DM, and average plant height (HEI). All genetic-statistical analyses were conducted using the Genes Program. The evaluated genotypes showed nonsignificant differences, indicating that, for the evaluated traits, these genotypes are genetically similar. Most genotypes presented linear effect of 2nd degree in the two assessment cuts, pointing they did not reach their maximum capacity of DMY in the time interval of the evaluated cuts. The maximum %DM achieved for the most genotypes occurred at about 9-mo age, while genotypes Vruckwona and BAG-86 at about 10-mo age. The maximum points for HEI trait were between 8 and 10-mo age. Increasing the cutting interval leads to increases in DM yield, with the 10-mo cutting age being the most efficient, resulting in higher yields.

ABSTRACT The long-term success of populations is dependent on individual reproductive success, and in general, increased population size and genetic diversity contribute to population maintenance, reducing the risk of local extinction. Interspecific hybridization has consequences that can vary according to the hybrids' fate, which can be strongly influenced by the reproductive capacity of hybrids and canonical individuals from the contact zone. We examined the reproductive biology and morphology of two closely related Petunia (Solanaceae) species and their interspecific hybrids from Serra do Sudeste, Southern Brazil, and we measured their reproductive success under controlled conditions based on seed production and germination from five pollination treatments. We found differences in self-compatibility degree among individuals, lineages, and pollination treatments based on high total seed production (> 204,000 seeds) and germination (630 seedlings evaluated). No correlation was observed between corolla colour and reproductive success or between floral morphological traits and compatibility. High self-compatibility and inter-lineage compatibility can explain the hybrid populations' maintenance and origin, favouring the two analysed species' introgression.

Abstract Introduction: The Technical Performance Score (TPS) was developed and subsequently refined at the Boston Children's Hospital. Our objective was to translate and validate its application in a developing country. Methods: The score was translated into the Portuguese language and approved by the TPS authors. Subsequently, we studied 1,030 surgeries from June 2018 to October 2020. TPS could not be assigned in 58 surgeries, and these were excluded. Surgical risk score was evaluated using Risk Adjustment in Congenital Heart Surgery (or RACHS-1). The impact of TPS on outcomes was studied using multivariable linear and logistic regression adjusting for important perioperative covariates. Results: Median age and weight were 2.2 (interquartile range [IQR] = 0.5-13) years and 10.8 (IQR = 5.6-40) kilograms, respectively. In-hospital mortality was 6.58% (n=64), and postoperative complications occurred in 19.7% (n=192) of the cases. TPS was categorized as 1 in 359 cases (37%), 2 in 464 (47.7%), and 3 in 149 (15.3%). Multivariable analysis identified TPS class 3 as a predictor of longer hospital stay (coefficient: 6.6; standard error: 2.2; P=0.003), higher number of complications (odds ratio [OR]: 1.84; 95% confidence interval [CI]: 1.1-3; P=0.01), and higher mortality (OR: 3.2; 95% CI: 1.4-7; P=0.004). Conclusion: TPS translated into the Portuguese language was validated and showed to be able to predict higher mortality, complication rate, and prolonged postoperative hospital stay in a high-volume Latin-American congenital heart surgery program. TPS is generalizable and can be used as an outcome assessment tool in resource diverse settings.

Abstract Introduction: Elevated neutrophil-lymphocyte ratio (NLR) has been associated with poorer outcomes in cyanotic patients undergoing single ventricle palliation. Little is known about this biomarker on patients with tetralogy of Fallot (TOF), the most common cyanotic congenital heart disease. Our objective is to study the impact of preoperative NLR on outcomes of TOF patients undergoing total repair. Methods: This retrospective study included 116 consecutive patients between January 2014 and December 2018. Preoperative NLR was measured from the last complete blood count test before the surgery. Using the cutoff value of 0.80, according to the receiver-operating characteristic (ROC) curve, the sample was divided into two groups (NLR < 0.80 and ≥ 0.80). The primary endpoint was hospital length of stay (LOS). Results: ROC curves showed that higher preoperative NLR was associated with longer hospital LOS, with an area under the curve of 0.801±0.040 (95% confidence interval 0.722 - 0.879; P<0.001). High preoperative NLR was also associated with long intensive care unit (ICU) LOS (P=0.035). Preoperative NLR predicted longer hospital LOS with a sensitivity of 63% and a specificity of 81.4%. Conclusion: Higher preoperative NLR was associated with long ICU and hospital LOS in patients undergoing TOF repair.

Resumo Introdução: A laringectomia supratraqueal tem sido descrita como um procedimento cirúrgico com objetivo de preservar a função da laringe (respiração, fonação e deglutição), sem prejuízo no controle oncológico locorregional, para câncer glótico ou supraglótico com extensão à subglote e/ou envolvimento da articulação cricoaritenóidea. A opção pela laringectomia supracricoide nesses casos poderia resultar em grande risco para margens de ressecção comprometidas. Objetivo: Determinar a segurança, viabilidade, adequação das margens cirúrgicas e os resultados da laringectomia supratraqueal para o câncer de laringe intermediário e avançado através da revisão dos resultados de três instituições distintas no Brasil. Método: Estudo retrospectivo, com análise dos prontuários de 29 pacientes submetidos à laringectomia supratraqueal, de outubro de 1997 a junho de 2017. O tipo de laringectomia realizada foi classificado de acordo com a classificação da Sociedade Laringológica Europeia para laringectomias horizontais. Foram avaliados os resultados precoces e tardios. As taxas de sobrevida (global, específica, livre de doença e livre de laringectomia total) foram calculadas. O tempo médio de seguimento foi 44 meses. Resultados: Dos 29 pacientes submetidos à laringectomia supratraqueal, 25 não tinham tratamento prévio. Um paciente (3,4%) teve margens comprometidas. Quatro pacientes (13,8%) recidivaram. Desses, três tiveram recidiva local e um apresentou recidiva regional. Cinco pacientes (17,2%) necessitaram de totalização da laringectomia, duas por ruptura da pexia e três por recidiva local. Quatro desses pacientes (80%) obtiveram sucesso na totalização. Quatro pacientes (13,8%) foram a óbito, dois por complicações pós-cirúrgicas e dois por recidiva. As sobrevidas global, específica, livre de doença e livre de laringectomia total em 5 anos foram, respectivamente, 82,1%; 88,2%; 83,0% e 80,2%. Conclusão: Pacientes selecionados com câncer intermediário e avançado de laringe podem ser beneficiados com laringectomia supratraqueal, que ofereceu sobrevida livre de laringectomia total e sobrevida específica de 80,2% e 88,2%, respectivamente.
Abstract Introduction: Supratracheal laryngectomy has been described as a surgical procedure for glottic or supraglottic cancer extending to the subglottic region and/or involving the cricoarytenoid joint, aiming to preserve laryngeal function (breathing, phonation and swallowing), without diminishing locoregional cancer control. The choice of supracricoid laryngectomy in these cases could result in a high risk of compromised resection margins. Objective: To determine the safety, viability, adequacy of surgical margins and the supratracheal laryngectomy results for intermediate and advanced laryngeal cancer by reviewing the results at three different institutions in Brazil. Methods: This is a retrospective study that analyzed the charts of 29 patients submitted to supratracheal laryngectomy from October 1997 to June 2017. The type of laryngectomy performed was classified according to the European Laryngological Society classification for horizontal laryngectomies. Early and late results were evaluated. Survival rates (overall, specific, disease-free and total laryngectomy-free survival) were calculated. The mean follow-up time was 44 months. Results: Of the 29 patients submitted to supratracheal laryngectomy, 25 had no previous treatment. One patient (3.4%) had compromised margins. Four patients (13.8%) had recurrence. Of these, three had local recurrence and one had regional recurrence. Five patients (17.2%) required a total laryngectomy, two due to ruptured pexy and three due to local recurrence. Four of these patients (80%) achieved a successful total procedure. Four patients (13.8%) died, two due to postoperative complications and two due to recurrence. Overall, specific, disease-free and total laryngectomy-free survival at 5 years were, respectively, 82.1%; 88.2%; 83.0% and 80.2%. Conclusion: Selected patients with intermediate and advanced laryngeal cancer may benefit from supratracheal laryngectomy, that resulted in total laryngectomy-free survival and specific survival of 80.2% and 88.2%, respectively.

RESUMO: A exposição inadequada do sistema reprodutor feminino a esteróides em períodos do desenvolvimento uterino pode inibir parcialmente o desenvolvimento das glândulas endometriais em cães. Entretanto, não se conhece os efeitos dos esteróides sobre a funcionalidade das glândulas formadas, bem como a possível ocorrência de fibrose endometrial. Objetivou-se avaliar a atividade secretória das glândulas endometriais de cadelas pré-púberes submetidas a protocolo de ablação parcial da adenogênese uterina. Foram utilizadas 16 fêmeas, sem-raça-definida, distribuídas nos grupos MPA (n=8), fêmeas que receberam aplicações de acetato de medroxiprogesterona a cada 3 semanas, e C (n=8), fêmeas controle não tratadas. Aos seis meses de idade, foi realizada ovariohisterectomia em todos os animais, e avaliados os cornos uterinos pelo exame histológico e de histoquímica. Para avaliar a intensidade de secreção (graus 1-4), foram utilizadas periodic acid-Schiff e alcian blue (AB) pH 2,5. Para a avaliação histológica foram utilizados tricrômico de Masson e azul de toluidina. Apenas marcações graus 1 e 2 foram observadas para PAS em ambos os grupos, sem diferença na intensidade de secreção uterina entre grupos com relação aos graus encontrados. Entretanto, o grupo MPA apresentou maior intensidade de secreção uterina em relação ao grupo C (p<0,05). Com relação ao AB pH 2,5, em ambos os grupos também foram encontradas apenas marcações de graus 1 e 2, sem diferença estatística entre grupos. Não foram observadas marcações para a coloração de tricrômico de Masson na região periglandular, em ambos os grupos. Foi observada maior quantidade de mastócitos presentes no útero na região do miométrio, em ambos os grupos. Conclui-se que cadelas pré-púberes com ablação parcial da adenogênese uterina apresentam mínima atividade secretória uterina, ausência de fibrose periglandular e maior presença de mastócitos no miométrio em relação ao endométrio.
ABSTRACT: Inadequate exposure of the female reproductive system to steroids in uterine developmental periods can partially inhibit the development of endometrial glands in dogs. However, the effects of steroids on the formed glands functionality remain unknown, as well as the possible occurrence of endometrial fibrosis. This study aimed to evaluate the secretory activity of endometrial glands in prebubertal female dogs submitted to a protocol of partial ablation of the uterine adenogenesis. Sixteen females of non-specific breed were distributed into two groups; MPA (n=8), females that received applications of medroxyprogesterone acetate every 3 weeks; and C (n=8) untreated control females. Ovariohysterectomy was performed in all animals at the age of 6 months and evaluated the uterine horns by histological and histochemistry exams. The secretion intensity (degrees 1-4) was evaluated using periodic acid-Schiff (PAS) and alcian blue (AB) pH 2.5. Histological evaluation was performed using Masson’s trichrome and toluidine blue. Only degree 1 and 2 marks for PAS were observed in both groups, with no difference of uterine secretion intensity between the groups regarding the degrees found. However, the MPA group revealed higher intensity of uterine secretion compared to group C (p<0.05). Staining with AB pH 2.5 also revealed only degree 1 and 2 marks in both groups, with no statistically significance between them. Masson’s trichrome staining revealed no marks in the periglandular region in both groups. A higher among of mast cells was observed in the myometrial region of the uterus in both groups. Prepubertal female dogs with partial ablation of the uterine adenogenesis present minimal uterine secretory activity, absence of periglandular fibrosis and increased presence of mast cells in the myometrium compared to endometrium.

What could be the fate of secondary contact zones between closely related plant species?

Abstract Interspecific hybridization has been fundamental in plant evolution. Nevertheless, the fate of hybrid zones throughout the generations remains poorly addressed. We analyzed a pair of recently diverged, interfertile, and sympatric Petunia species to ask what fate the interspecific hybrid population has met over time. We analyzed the genetic diversity in two generations from two contact sites and evaluated the effect of introgression. To do this, we collected all adult plants from the contact zones, including canonicals and intermediary colored individuals, and compared them with purebred representatives of both species based on seven highly informative microsatellite loci. We compared the genetic diversity observed in the contact zones with what is seen in isolated populations of each species, considering two generations of these annual species. Our results have confirmed the genetic differentiation between the species and the hybrid origin of the majority of the intermediary colored individuals. We also observed a differentiation related to genetic variability and inbreeding levels among the populations. Over time, there were no significant differences per site related to genetic diversity or phenotype composition. We found two stable populations kept by high inbreeding and backcross rates that influence the genetic diversity of their parental species through introgression.

Characterization of the complete plastid genome of Butia eriospatha (Arecaceae)

Abstract Butia eriospatha is an endemic palm species from the Atlantic Rainforest in Brazil, a biodiversity hotspot. This species is currently listed in the IUCN red list as vulnerable and lacks specific plastid markers for population genetics studies. In addition, the evolutionary relationship within the genus Butia is not yet well resolved. Here, we sequenced and characterized the complete plastid genome (plastome) sequence of B. eriospatha. The complete plastome sequence is 154,048 bp in length, with the typical quadripartite structure. This plastome length and genes content is consistent with other six species from tribe Cocoseae. However, the Inverted Repeat (IR) borders show some variation among the analyzed species from this tribe. Species from the Bactridinae (Astrocaryum and Acrocomia) and Elaeidinae (Elaeis) subtribes present the rps19 gene completely duplicated in the IR region. In contrast, all plastomes sequenced from the subtribe Attaleinae (Butia, Cocos, Syagrus) present one complete CDS of rps19 and one partial copy of rps19. The difference in the IR/LSC junctions between Attaleinae and the sister clades Bactridinae + Elaeidinae might be considered an evolutionary signal and the plastome sequence of B. eriopatha may be used in future studies of population genetics and phylogeny.

ABSTRACT Interspecific hybridization has several consequences for parental species, from blurring species limits to the emergence of new taxa. Petunia axillaris and P. exserta occur in sympatry in southernmost Brazil and naturally hybridize despite their different pollination syndromes and habitat requirements. We employed genetic and morphological analyses to characterize two contact zones between the species with the aim of determining the effect of interspecific hybridization. Microsatellite loci and a morphometric evaluation of the corolla shape were used to classify individuals based on their origin as pure parental or hybrids. Corolla color was used to classify individuals a priori (white, red or intermediate, for P. axillaris, P. exserta or hybrid, respectively). Corolla color was found to be a good indicator of the genetic component of each species and their hybrid, while the shape of the corolla did not always correspond to genetic origin. Hybridization increased the variability, and introgression occurred in both directions in this system.

RESUMO: O objetivo deste estudo foi avaliar, por meio de simulação de dados, o impacto das restrições no número máximo de irmãos completos e meios-irmãos selecionados para machos e fêmeas no nível de endogamia e ganho genético do rebanho. Os dados originais são provenientes das populações reais A e B, compostas por suínos da raça Pietrain e Landrace, respectivamente. Para gerar as populações simuladas, foi desenvolvido um simulador em linguagem Fortran utilizando as (co)variâncias dos valores genéticos e as taxas produtivas e reprodutivas obtidas das populações A e B. Dois arquivos de dados foram criados. O primeiro continha o pedigree dos 10 anos anteriores, com 21.906 e 251.343 animais nas populações A e B, respectivamente. O segundo incluiu os valores genéticos para idade para atingir 110 Kg de peso vivo, espessura de toucinho e conversão alimentar, para ambas as populações; profundidade do músculo longissimus dorsi, apenas para a população A; e número de leitões vivos no 5º dia de vida por parto, apenas para a população B. Três cenários foram simulados com dez gerações, variando as restrições quanto ao número de irmãos completos e meios-irmãos selecionados para machos e fêmeas, com 30 repetições por geração e cenário. Independentemente da estratégia de acasalamento utilizada em um núcleo de produção fechada, há aumento nos níveis de endogamia. Aumentos de endogamia são maiores em populações de menor tamanho efetivo. Restrições ao número de irmãos completos e meios-irmãos selecionados são eficazes na redução de incrementos na endogamia. A restrição de no máximo dois irmãos completos, três meios-irmãos para machos e três irmãs completas para fêmeas fornece os maiores ganhos genéticos.
ABSTRACT: The objective of this study was to evaluate, through data simulation, the impact of restrictions on the maximum number of full- and half-sibs selected for males and females on the level of inbreeding and genetic gain of the herd. Data came from real populations A and B, composed of Pietrain and Landrace breed pigs, respectively. To generate the simulated populations, a Fortran-language simulator was developed using the (co)variances of the breeding values and the productive and reproductive rates obtained from populations A and B. Two data files were created. The first contained the pedigree of the previous 10 years, with 21,906 and 251,343 animals in populations A and B, respectively. The second included breeding values for age to reach 110 Kg body weight, backfat thickness, and feed conversion, for both populations; longissimus dorsi muscle depth, for population A only; and number of live piglets at the 5th day of life per farrowing, for population B only. Three scenarios were simulated with ten generations by varying the restrictions on the number of full- and half-sibs selected for males and females, with 30 replicates per generation and scenario. Regardless of the mating strategy used in a closed production unit, there is an increase in inbreeding levels. Inbreeding increases are larger in populations of smaller effective size. Restrictions on the number of full- and half-sibs selected are effective in reducing increments in inbreeding. Restriction to a maximum of two full-sibs and three half-sibs for males and three full sisters for females provided the highest genetic gains.

Due to the economic potential of camu-camu, considering its high concentration of vitamin C, the aim of this work was to evaluate the quality and genetic variability seven accessions of camu-camu conserved in genebank (BAG) of Embrapa Amazônia Oriental, in the state of Pará, Amazon region, Brazil. The fruits of camucamuzeiro were analyzed for physicochemical characterization (standard methodologies) and mineral composition. The data were subjected to multivariate statistical analysis, using the techniques of cluster analysis and principal component analysis. The formation of different groups for each genotype, which shows the genetic variability and the dissimilarity of the genotypes in the species data, may be used to guide the selection of promising genotypes to enrich the programs of genetic improvement of the camucamuzeiro. Ascorbic acid showed levels above 1000 mg 100 g-1, however, genotypes 4, 1 and 2 show the highest potential and the most promising nutritional capacity, but genotype 4 showed good characteristics for the moisture, acidity, carbohydrates, Cu and Zn and differs totally from others about the total soluble solids (TSS) and flavon-3-ol, features that make it the most promising genotype. It was possible to separate the seven different genotypes using multivariate analysis (hierarchical cluster analysis-HCA and principal component analysis-PCA).

The present study aimed to investigate the presence of compounds with antitumor activity in the plant Caesalpinia pluviosa var. peltophoroides. From bioactivity guided studies it was possible to isolate a new biflavonoid, named caesalpinioflavone, whose chemical structure was determined by spectroscopic (1H and 13C nucler magnetic ressonace, homonuclear correlation spectroscopy and heteronuclear multiple-bond correlation spectroscopy) and spectrometric (high resolution electrospray ionization mass) methods. According to in vitro assays, caesalpinioflavone was effective in reducing the cell viability of tumor cell lines A549, MCF7, Hst578T and HTC. This effect was consequent of cell cycle arrest in G1/S transition (A549 and MCF7) and cytotoxic activity (Hs578T and HTC). Taken together, these data indicate that caesalpinioflavone has a promising antitumor activity.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Sodium bicarbonate as prevention of metabolic acidosis in sheep submitted to experimental ruminal acidosis

O objetivo deste estudo foi avaliar o efeito preventivo do bicarbonato de sódio sobre a acidose sistêmica em decorrência da acidose ruminal, a qual foi induzida pelo fornecimento de concentrado após jejum prolongado. Foram utilizados 14 ovinos, divididos em três grupos experimentais: grupo controle (Cg), contendo quatro ovinos, submetidos a jejum sem desenvolvimento de acidose ruminal; grupo não tratado (NTg), contendo cinco ovinos submetidos a acidose ruminal sem tratamento preventivo; e grupo tratado (Tg), contendo cinco ovinos, submetidos a acidose ruminal e tratados preventivamente com bicarbonato de sódio. Foram realizadas avaliações do pH ruminal e hemogasometria arterial, durante 48 horas após o fornecimento do concentrado. Houve redução do pH ruminal em todos os grupos, sendo que o Cg apresentou a redução apenas às 24 horas. Notou-se redução do pH arterial, bicarbonato e excesso de base em todos os grupos, indicando acidose metabólica sistêmica; no entanto, o NTg apresentou o quadro mais grave. Conclui-se que o bicarbonato de sódio possui efeito preventivo da acidose metabólica sistêmica, reduzindo a sua gravidade em ovinos submetidos à acidose ruminal.
The aim of this study was to evaluate the preventive effect of sodium bicarbonate on systemic acidosis due to ruminal acidosis, which was induced by ingestion of concentrate after prolonged fasting. Fourteen sheep were divided into three experimental groups: control group (Cg), with four sheep, submitted to fasting without development of ruminal acidosis; no-treated group (NTg), with five sheep with rumen acidosis without preventive treatment; and treated group (Tg), with five sheep with rumen acidosis and preventively treated with sodium bicarbonate. Assessments of ruminal pH and arterial hemogasometry were performed for 48 hours after ingestion of the concentrate. There was a reduction in the ruminal pH in all groups, whereas the Cg showed a reduction only after 24 hours. A reduction in the arterial pH, bicarbonate and base excess in all groups was also noted, indicating systemic metabolic acidosis, but the NTg presented the greatest alteration. It is concluded that sodium bicarbonate prevents systemic metabolic acidosis, reducing its severity in sheep subjected to ruminal acidosis.
